Queen Marie of Denmark was the daughter of Landgrave Carl of Hesse-Kassel (1744–1836), governor of Schleswig-Holstein, and Landgrave Louise of Hesse-Kassel, née Princess of Denmark as the daughter of King Frederik V of Denmark. Queen Marie was thus the sister of Duchess Louise of Glücksburg, married to Duke Wilhelm of Glücksborg. Therefore, the Royal couple were aunt and uncle to the Ducal couple's ten children, including King Christian IX of Denmark, Duke Carl of Glücksburg, Duke Friedrich of Glücksburg, Duchess Friederike of Anhalt-Bernburg and Prince Hans of Glücksburg. The Royal couple took special care of their nephew, Christian, who was proclaimed King of Denmark in 1863.
