Princess Henriette was the daughter of Duke Friedrich (1757–1816) and Duchess Friederike of Schleswig-Holstein-Sonderburg-Beck (1757–1827). She was married in 1803 to Prince Friedrich Ferdinand of Anhalt-Köthen-Pless (1769–1830). Princess Henriette was the sister of Duke Wilhelm of Schleswig-Holstein-Sonderburg-Glücksburg (1785–1831), married to Princess Louise of Hesse-Kassel. This latter couple became the parents of, among others, King Christian of Denmark Duke Carl of Glücksborg, Duke Friedrich of Glücksborg and Duchess Frederikke of Anhalt-Bernburg. This group of siblings were thus nephews and nieces of Princess Henriette of Schleswig-Holstein-Sonderburg-Beck.
